Abstract

Protein-protein interaction (PPI) inhibitor discovery has proven difficult due to the large surface area and dynamic interfaces.To detect these interactions, the trace amounts of organic fluorine in biology has led scientists to benefit from this bioorthogonality and use fluorine to visualize biological phenomenon by magnetic resonance. In the first part of the talk, I will described our efforts to facilitate the early lead discovery rate of small molecules for PPIs using a rapid protein-based 19F NMR method for detecting protein-ligand interactions called PrOF NMR. In the second part of the talk, I will describe our molecular design efforts for creating new peptide-based 19F MRI agents.I will present our research design strategy for developing highly fluorinated polyamides with tunable biological and environmental stability.
